Anti-CD45抗體說明書,白細(xì)胞共同抗原CD45抗體產(chǎn)品介紹：The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains an extracellular domain, a single transmembrane segment and two tandem intracytoplasmic catalytic domains, and thus belongs to receptor type PTP. This gene is specifically expressed in hematopoietic cells. This PTP has been shown to be an essential regulator of T- and B-cell antigen receptor signaling. It functions through either direct interaction with components of the antigen receptor complexes, or by activating various Src family kinases required for the antigen receptor signaling. This PTP also suppresses JAK kinases, and thus functions as a regulator of cytokine receptor signaling. Four alternatively spliced transcripts variants of this gene, which encode distinct isoforms, have been reported.

福爾馬林固定的組織有可能破壞了原來組織的抗原結(jié)構(gòu)，蛋白多肽發(fā)生交聯(lián)，導(dǎo)致部分抗原表位封閉，結(jié)合位點減少，所以需要抗原修復(fù)，以暴露原來的抗原表位，達(dá)到充分顯露抗原，以不出現(xiàn)明顯脫片現(xiàn)象為佳。

抗原修復(fù)的幾種常用方法（供參考）
1、 微波爐加熱：
在微波爐里加熱0.01*緩沖液（pH6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復(fù)1-2次。根據(jù)玻片情況可參考采用微波加熱“3-5-3法”3分鐘溫火沸騰后靜止5分鐘，再溫火沸騰3分鐘即可。
適用的抗原：來源于人、大鼠、小鼠的組織標(biāo)本；來源于其他動物種屬的組織標(biāo)本。
2、 沸熱修復(fù)：
電爐或水浴鍋加熱0.01M*緩沖液（pH6.0）至95℃左右，放入組織芯片加熱10-15分鐘。
3、 高壓熱修復(fù)：
在沸水中加入EDTA（pH8.0）或0.01m*緩沖溶液（pH6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色架上，緩慢加壓，使玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會升起來。10分鐘后除去熱源，置入涼水中，當(dāng)小閥門沉下去后打開蓋子。此方法適用于較難檢測或核抗原的抗原修復(fù)。（骨及軟骨組織的標(biāo)本選擇較溫和的微波加熱修復(fù)） 。
4、 酶消化方法：
常用0.1%*和0.4%*液。*使用前預(yù)熱37℃，消化時間約為5-30分鐘。
適用于被固定遮蔽的抗原：包括 Collagen.GFAP. Complement.Cytokeratin.c-erB-2.LCA.LN.等。

抗體修復(fù)方式:
修復(fù)液：0.01M 枸櫞酸（pH6.0）或者0.05M EDTA（pH8.0）
抗體修復(fù)方法：
方法1：沸水浴修復(fù)，將盛有修復(fù)液和玻片的燒杯置于沸水浴環(huán)境，保持外部沸騰狀態(tài)15min，自然冷卻至室溫；
方法2：微波修復(fù)，將盛有修復(fù)液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。
