Anti-PCDH20抗體,原鈣粘附蛋白20抗體產(chǎn)品介紹：As a subfamily of the cadherin superfamily, protocadherins are cadherin-like cell adhesion proteins that contain up to seven extracellular domains and are predominantly expressed in the nervous system. Importantly, the adhesion mechanism of protocadherins is distinct from classic cadherins. Through inactivation or overexpression, several protocadherins have been implicated in a variety of cancers. Protocadherin-20 (PCDH20), also known as protocadherin-13, is a 924 amino acid protein containing 6 cadherin domains and potentially functioning as a calcium-dependent cell-adhesion protein. In non-small cell lung cancer cell lines, a homozygous loss of PCDH20 was identified through either deletion of one allele and methylation of the other or methylation of both alleles. Hypermethylation of PCDH20 is associated with worse prognosis and clinical outcome, suggesting that PCDH20 may function as a tumor suppressor.

抗原修復(fù)的幾種常用方法（供參考）
1、 微波爐加熱：
在微波爐里加熱0.01*緩沖液（pH6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復(fù)1-2次。根據(jù)玻片情況可參考采用微波加熱“3-5-3法”3分鐘溫火沸騰后靜止5分鐘，再溫火沸騰3分鐘即可。
適用的抗原：來(lái)源于人、大鼠、小鼠的組織標(biāo)本；來(lái)源于其他動(dòng)物種屬的組織標(biāo)本。
2、 沸熱修復(fù)：
電爐或水浴鍋加熱0.01M*緩沖液（pH6.0）至95℃左右，放入組織芯片加熱10-15分鐘。
3、 高壓熱修復(fù)：
在沸水中加入EDTA（pH8.0）或0.01m*緩沖溶液（pH6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色架上，緩慢加壓，使玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會(huì)升起來(lái)。10分鐘后除去熱源，置入涼水中，當(dāng)小閥門沉下去后打開(kāi)蓋子。此方法適用于較難檢測(cè)或核抗原的抗原修復(fù)。（骨及軟骨組織的標(biāo)本選擇較溫和的微波加熱修復(fù)） 。
4、 酶消化方法：
常用0.1%*和0.4%*液。*使用前預(yù)熱37℃，消化時(shí)間約為5-30分鐘。
適用于被固定遮蔽的抗原：包括 Collagen.GFAP. Complement.Cytokeratin.c-erB-2.LCA.LN.等。

修復(fù)液：0.01M 枸櫞酸（pH6.0）或者0.05M EDTA（pH8.0）
抗體修復(fù)方法：
方法1：沸水浴修復(fù)，將盛有修復(fù)液和玻片的燒杯置于沸水浴環(huán)境，保持外部沸騰狀態(tài)15min，自然冷卻至室溫；
方法2：微波修復(fù)，將盛有修復(fù)液和玻片的燒杯置于微波爐中，高火5min，?；?min，中火5min，自然冷卻至室溫。
