Progesterone measurement
The steroids from 0.05?0.1 g of each thoroughly mixed yolk sample were extracted with 1 mL diethyl ether and purified by a precipitation with 1 mL 95 % ethanol to remove excess lipids. Progesterone levels in the extracts were then measured using goose progesterone ELISA kit (Shanghai Huding Biotechnology Co., Ltd., Shanghai, China) according to the manufacturer’s instruction, and total progesterone levels in each follicle were calculated as [(the total levels of progesterone in the extract ÷ the weight of yolk subjected to extraction) × the total weight of yolk isolated from this follicle] and were expressed as pmol/follicle. Levels of progesterone in culture media were also measured using goose progesterone ELISA kit (Shanghai Huding Biotechnology Co., Ltd., Shanghai, China). Serial dilutions of media samples showed good linearity (differed by 15 % or less) in the calculated concentrations of progesterone, and the detection limit of progesterone is 7 pmol/L. Relative progesterone level was expressed as the fold difference vs. cells treated with the siRNA-NC or the pEGFP-N1 empty plasmid
